What PC do you have?
Which OS, servicepack?
How many COM ports are defined?
Which version of IDE?
Have you tried the UNO on another PC to see if the same message pops up?

,Thanks  for reply,  last question first, it works OK on another computer, I just tried it. Hooray !

The laptop I want to use it on that doesn't work is;
HP Presario  CQ57   Notebook  PC,      Windows 7  Home Premium,  64 bit , Service pack 1

How do I define the COM ports and find out the IDE version ?

Thanks again,
