I want to be able to use the hardware serial lines with a device, but only when I drive pin 2 high. Otherwise, I want the serial device to be effectively disconnected from the Arduino serial pins. Is it possible to do this just using transistors?

Yes you could do it with transistors but it is much easier using a 4051, see:-
