I planning to power Arduino 2560 and Arduino WiFi Shield from 12 V car battery.
Does it works fine ?

Yes, if the battery is not connected to a car. If the battery is connected to a car you should anticipate a lot of noise in the power lines.

Best to use a separate voltage regulator from the vehicle battery to 7 to 9 volts.  Protects against the spikes and surges on the vehicle.
