Can anyone explain this error message and what I can do about it?

option enabled in File -> Preferences.

Do you have the correct serial port selected?

Please try updating to Arduino IDE 1.6.11. There is a known issue with 1.6.10 that causes this error when you start the IDE by opening a sketch file.

Let us know if that does/doesn't fix the issue.
