Can I use an SD and a Micro SD with the same reader?

Hello,
I need to use two SD Cards in the design that I have been working on, one for the primary storage and another for backup. I have this module which can support both SD and MicroSD. My question is rather than use another module for the backup SD, could I use both slots on the module for my needs?

If the board only has a single CS connection I would guess that the two different card formats are simply alternatives.
