The Pocket Programmer's connector has pin labels associated with SPI.
Will it work with the Mini with the following connections?
Pocket --> Mini
MOSI --> Rx
MISO --> Tx
VCC --> +5
GND --> GND
RST --> R

Nope. The Programmer does not speak TTL serial (At least in that way).
You could hook it up normally (MOSI->MOSI, MISO->MISO, SCK->SCK, etc) and upload a sketch and over-write the bootloader. If you want to put the bootloader back on, you will have to re-burn it.

As long as it is Linux, MacOS or WinXP, no problem. If it is WinCE, the IDE will not work.
