But if I choose to use the T-15A standard USB cable connecting with the Arduino Mega with a USB host, do I still need the converter ? And should I still use the SoftwareSerial to send commands and receive responses if the two devices are connected by USB?

I am sorry, I don't really understand the question.

If I were you, I would use UNO + SoftwareSerial, because:

  1. Much simpler connection
  2. It seems the protocol of the light meter is quite simple, so no need to use converter (99% of my projects that involve connecting RS232 devices don't require converter)

What about the wiring between the light meter to arduino?
If you use: SoftwareSerial mySerial(7,8,true);
Then connect the RX line of lightmeter to pin 8 and TX line to pin 7. Gnd of lightmeter to Gnd of arduino
