Check all your settings under the tools heading to make sure you have you have the correct board, programmer and COM port selected. The IDE has 10 goes at uploading then gives up. Incorrect settings are the usual cause before suspecting something else. It's quite easy to change settings accidentally just by flicking your cursor over the menu items.
Is this a genuine Arduino Uno with a 16u secondary processor for USB to serial?. Some cloned boards have CH340 chips and don't use the default processor setting. You need to use setting "old bootloader"
