I installed the Arduino mega and 2x16 OLED into my car with a 5 volt power supply directly to 5 volt connections on the proto shield. Is it damaging to the mega if I connected my USB from the laptop along with the 5 volt strait to board?

No. The PC may not be too happy - there will only be the 500mA resettable polyfuse between the USB from the PC and the 5V coming in from the header pin.
