Access denied on serial port typically indicates that another program is using the serial port. If you can't find any application you were using that's doing that, and it persists after reboot, check if you have any software related to modems running in the background - some modem software assumes that any serial port connected is a modem, and tries to talk to it as if it was one - in the process blocking any other program from using the serial port.
