Tx lights up when the Arduino board sends information, typically by Serial.print()ing.
Rx lights up when the Arduino board receives information, typically because you type something in the input bar area of the serial monitor. And hit return.
So if you have the serial window open those LEDs do not have much diagnostic value to add.
