I’m looking to ground roughly 50 dupont wires ?

Like a "Ground Shield" with 50-60 points?

You can make something like this with an IDC connector.

Correct.

A bench vice is used to close the connector.

I've thought of laying one out in 'Eagle' (because absurdities appeal to me).

Or something like this:

Connect all the pins to ground on the underside of the board and clamp the duPont pins into the block's pockets

I buy them by the dozen. the fact that they are marked for VCC and I2C does not compel you to use them for VCC and I2C

same thing different way: the black and red lines are all tied together
