I was curious about the feasibility of an Arduino Due running two sets of codes simultaneously. For instance, void setup/void loop while also running 3d printer g-code? I intend to use it to communicate and execute commands from Nextion (using g-code designations) like turning on relays while running 3d printer jobs. Any ideas?

It should be possible. But you can only upload one program to an Arduino so you would have to modify the 3D printer program to include the extra functionality - not a task for the faint-hearted or the inexperienced.
